Anti-BMP4 produced in rabbit, affinity isolated antibody Catalog Number SAB4200556 Product Description Anti-BMP4 is produced in rabbit using as immunogen a synthetic peptide corresponding to an internal sequence of human BMP4 (GeneID: 652), conjugated to KLH.

The corresponding sequence is highly conserved (single amino acid substitution) in rat and mouse BMP4. The antibody is affinity-purified using the immunizing peptide immobilized on agarose. Anti-BMP4 specifically recognizes human BMP4. The antibody may be used in several immunochemical techniques including immunoblotting (~55 kDa), immunoprecipitation and immunohistochemistry. Detection of the BMP4 band by immunoblotting is specifically inhibited by the BMP4 immunizing peptide. Bone morphogenetic proteins (BMPs) are members of the TGF-β superfamily. BMPs were originally identified as growth factors that can induce bone and cartilage formation.1,2 More than

30 members of the BMPs family have been identified which are involved in a large variety of functions during development and in cancer. BMPs are synthesized as precursor molecules, followed by enzymatic cleavage and release as active ligands. BMPs exert their biological function by binding to Ser/Thr kinase receptors including ALK2, BMPR-IA/ALK3, BMPR-IB/ALK6 and BMPR-II, and to phosphorylated Smads, mainly Smad1, Smad5 and Smad8.2 BMP4 (known as ZYME, BMP2B) regulates cell proliferation, differentiation, apoptosis and cell fate determination during development. Specifically, BMP4 has been shown to regulate mesodermal cell commitment to the hematopoietic cell lineage during early embryogenesis.

3 BMPs are expressed in localized patterns during embryonic mouse brain development.1 BMP4 is crucial for development since knockout mice die early in embryogenesis. Overexpression of BMP4 has been reported in several cancers, including melanoma, hepatocellular carcinoma, adenocarcinoma and colorectal cancer and is frequently accompanied by an enhanced cell migratory and invasive potential of the tumors.2,4-6 Reagent Supplied as a solution in 0.01 M phosphate buffered saline, pH 7.4, containing
