电话:400-6721-600 Human Growth Hormone/GH ELISA Kit DESCREPTION Growth hormone (GH), also known as somatotropin, is a peptide hormone that stimulates growth, cell reproduction, and cell regeneration in humans and other animals. It is thus important in human development. GH is a stress hormone that raises the concentration of glucose and free fatty acids. It also stimulates production of IGF-1. The most common disease of GH excess is a pituitary tumor composed of somatotroph cells of the anterior pituitary. The effects of GH deficiency vary depending on the age at which they occur. In children, growth failure and short stature are the major manifestations of GH deficiency, with common causes including genetic conditions and congenital malformations. It can also cause delayed sexual maturity. In adults, deficiency is rare, with the most common cause a pituitary adenoma, and others including a continuation of a childhood problem, other structural lesions or trauma, and very rarely idiopathic GHD. PRINCIPLE OF THE ASSAY This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for human Growth Hormone has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and Growth Hormone present is bound by the immobilized antibody. After washing away any unbound substances, a biotin-linked detect antibody specific for Growth Hormone is added to the wells. Following a wash to remove any unbound antibody-biotin reagent, streptavidin-HRP is added. After washing, substrate solution is added to the wells and color develops in proportion to the amount of Growth Hormone bound in the initial step. The color development is stopped and the intensity of the color is measured. LIMITATIONS OF THE KIT FOR RESEARCH USE ONLY. NOT FOR USE IN DIAGNOSTIC PROCEDURES. The kit should not be used beyond the expiration date on the kit label. Do not mix or substitute reagents with those from other lots or sources. If samples generate values higher than the highest standard, dilute the samples with Assay Buffer and repeat the assay. If cell culture supernate samples require large dilution, perform an intermediate dilution in culture medium. Any variation in standard dilution, operator, pipetting technique, washing techniques, incubation time or temperature, and kit age can cause variation in binding. This assay is designed to eliminate interference by factors present in biological samples.
